Caroverine hydrochloride:

Caroverine hydrochloride, also known by its popular brand names Spasmium, Tinnex, Tinnicar, Spasmium, and Tinnitin. Is a medication belonging to a group of pharmaceutical products which serve as muscle relaxants, originally used to relieve spasms in smooth muscles such as the intestines, arteries, and other similar organs. The use of caroverine hydrochloride has since expanded, with caroverine hydrochloride manufacturers and caroverine hydrochloride suppliers producing it for sale to distributors in Indian, Austrian, and Switzerland’s marketplace as a treatment for tinnitus. Currently, caroverine hydrochloride can be found both for sale and wholesale on the global marketplace at various price points, with its specific uses and cost being dependent on the country that it is licenced for use in. For example, it has not yet been approved for any treatment by the FDA in the United States.
Caroverine hydrochloride was initially discovered in Austria in the 1950s by the drugs manufacturing company Phafag AG. In 1972 its INN name of ‘caroverine’ was proposed and subsequent clinical trials for its use in the treatment of tinnitus occurred in 1997 and 2005. With Phafag licensing the rights for caroverine to the Indian company, Lincoln Pharmaceuticals, to develop the drug for tinnitus in 2010.

Method of action of caroverine hydrochloride:

Pharmacologically, caroverine hydrochloride is a nonspecific calcium channel blocker, which acts as an antagonist to both non-NMDA and NMDA glutamate receptors. This improves mechanosensitivity, mechanotransduction, and acts as an otoneuroprotective agent. It serves this function by acting as an N-type calcium channel blocker, competitive AMPA receptor antagonist, and non-competitive NMDA receptor antagonist. Therefore, when superfluous glutamate binds on to NMDA receptors, it opens, enabling calcium and sodium ions to enter the neuron. This disrupts the ionic balance and causes a subsequent depolarization state of the cell. This pathological depolarization state is reverted to the physiological polarization state by the antagonistic property of caroverine. Thus, mediating its observed effects.

Form and dosage of caroverine hydrochloride:

As with all drugs, the final packaging is dependent on the manufacturers and suppliers, as well as the distributors and intended marketplace. However, caroverine hydrochloride typically comes in tablet, film-coated tablet, vials, solution for injection, and capsule form. It is a prescription medication, as such, it cannot be taken without the advice of a medical professional. For the oral treatment of smooth muscle spasms, 20-40 mg 3-4 times daily to a maximum dose of 200 mg/day is typically prescribed. With parenteral treatment commonly being IV/IM of 40-80 mg daily, to a maximum of 200 mg/day.
