SOLIFENACIN SUCCINATE
Solifenacin Succinate is used to treat the symptoms of urge incontinence and or increased
frequency of urination and urgency as faced by patients with overactive bladder syndrome, a
condition in which the bladder muscles contract uncontrollably, causing frequent urination, and
those who lower or lost the capacity to control urination or the urge of it. It also treats
neurogenic detrusor overactivity, a form of bladder dysfunction caused by neurological
impairment, in children ages two years and older. Solifenacin Succinate is a muscarinic
antagonist.
Patients with NDO have an overactive bladder wall muscle, which should normally relax to allow
storage of urine. An overactive bladder wall muscle causes sporadic bladder muscle
contraction, thus building pressure on the bladder and decreases the volume of urine it can
hold. Spontaneous bladder muscle contractions also cause unexpected and frequent leakage of
urine with the immediate urge to urinate, urinating more often than normal, and loss of bladder
control.
Solifenacin succinate is sold under the brand name VESIcare, Bispec, Solicept among others.
Astellas Pharma Inc. filed the first known patents for the Process of producing solifenacin or its
salt in 2004. Aurobindo Pharma Ltd filed for a patent with USFDA in the year 2011 and was
granted the patent in 2014.
Dosage and Administration
Solifenacin succinate is available as tablets of 5mg. the usual recommended adult dose is 5 mg,
taken once a day. but if the patient tolerates the 5 mg dose, it can be increased by 10 mg. it
should be taken with water and swallowed whole and can be taken with or without meals.
The price of Solifenacin succinate
In the USA, the consumer can buy a common generic version of Solifenacin succinate for
around $14.48 for 30 tablets of 10mg, however, the average retail price is $342.51 for the same
supply. In Canada, consumers can buy a generic brand of 10 mg tablets for $101.99 for a
supply of 100 tablets.
How does it work?
Solifenacin antagonizes cholinergic receptor competitively and selectively for the M3 receptor
subtype. Contraction of smooth muscle is achieved by binding acetylcholine to these receptors,
particularly M3, which plays a critical role in the process. Solifenacin prevents acetylcholine from
binding to these receptors, which results in reducing the smooth muscle tone in the bladder.
This eventually allows the bladder to retain larger volumes of urine and reducing the number of
micturition, urgency, and incontinence episodes.
