Busulfan:

Busulfan, also known under the popular company brand names of Myleran (GlaxoSmithKline) and Busulfex IV (Otsuka America Pharmaceutical Inc), is a medication used in cancer chemotherapy treatments. Busulfan was originally approved by the FDA in the United States for the treatment of chronic myeloid leukaemia in 1999. In which it remained the mainstay chemotherapeutic treatment until it was predominantly displaced by the drug imatinib. Busulfan, however, still is used to a degree as a result of the drug's relatively low cost. This has made it attractive to busulfan manufacturers and busulfan suppliers, as it has entered the generic drugs space. Currently, it can frequently be found on pharmaceutical for sale list of distributors across the global marketplace, where it is available to order wholesale at a variety of cost and price points.

Method of action of busulfan:

Busulfan belongs to a group of drugs termed alkylating agents. These contain 2 labile methanesulfonate groups attached to opposite ends of a 4-carbon alkyl chain. Post hydrolysation, the methanesulfonate groups are released and carbonium ions are produced. This causes the acylation of DNA, which intern results in interference of DNA replication and RNA transcription. Culminating in disruption of nucleic acid function. This mechanism of action is enabled through alkylation which produces guanine-adenine intrastrand crosslinks. These crosslinks occur through an SN2 reaction, with guanine N7 mediating a nucleophilic attack on the carbon adjacent to the mesylate leaving group. This causes damage to the cellular DNA that can not be repaired by cellular machinery, thus triggering the cell to undergo apoptosis.

Form and dosage of busulfan:

The final form and packaging of busulfan are dependent on the manufacturers and suppliers, as well as the specific distributors and order. However, busulfan can typically be found for wholesale in forms including a solution for infusion, tablets, solution for injection, ampules, and vials. The typical dose administered is highly dependent on the patient and the specific underlying indication. As such it will vary on a patient by patient basis. For further information please refer to the manufacturers’ literature or contact a relevant healthcare provider.
