Niclosamide:

Niclosamide is a medication commonly known by its popular brand name Niclocide and is predominantly used to treat tapeworm infestations such as diphyllobothriasis, hymenolepiasis, and taeniasis. It has however been shown not to be effective against other worm infection types including pinworms or roundworms. Originally discovered in 1958 and subsequently listed for sale and wholesale on the global marketplace by the pharmaceutical products company Bayer, under the brand name Niclocide. It was voluntarily withdrawn from the marketplace in 1996, a decision influenced by the termination of the drugs patent and the emergence of generic niclosamide manufacturers and niclosamide suppliers, capable of listing niclosamide for sale to distributors at lower price points. Currently, it is on the World Health Organization's List of Essential Medicines, indicating niclosamide as an effective and cost efficient medication recommended for use in health systems around the globe.

Method of action of niclosamide:

Niclosamide is an antihelminth which functions through inducing the death of the parasitic worm upon contact. This is mediated through the uncoupling of oxidative phosphorylation or modulation of ATPase activity. This effectively disrupts the electron transport chain to ATP synthase, a crucial metabolic pathway necessary for the manufacturing of adenosine triphosphate (ATP). ATP is the essential molecule that supplies energy for metabolism, as such, lack of it results in organism death. Once the parasitic worm is killed or stunned by niclosamide, it passes normally through the digestive tract where it is digested and destroyed. Additional evidence also indicates that niclosamide could also function as molluscicide by binding to and damaging worm DNA.

Form and dosage of niclosamide:

Although the packaging is dependent on the manufacturers and distributors, niclosamide typically comes in tablet, chewable tablet, and film-coated tablet form. These are for oral consumption with water, and with or without food. Dosages are dependent on the prescribing medical professional and the underlying parasite being treated. With manufacturers recommending that patients over 6 years of age take 2 g in a single dose to treat T. saginata, T. solium and D. latum infections. With those under the age of 6 years being given 500 mg to 1 g in a single dose, depending on the size of the individual. For infections involving H. nana, recommended dosages for those over 6 years are an initial dose of 2 g followed by a 6-day regime of 1 g once a day, with values similarly lowered for young children.
